- Glutathione S-transferases are a family of enzymes that play an important role in detoxification by catalyzing the conjugation of many hydrophobic and electrophilic compounds with reduced glutathione. Based on their biochemical, immunologic, and structural properties, the mammalian cytosolic GSTs are divided into several classes, including alpha, mu, kappa, theta, pi, omega, and zeta. In addition, there is a class of microsomal GSTs. Each class is encoded by a single gene or a gene family. Promoter hypermethylation is an alternative pathway for gene silencing in neoplastic cells and a promising cancer detection marker. Quantitative methylation-specific PCR (QMSP) of the GSTP1 promoter has demonstrated near perfect specificity for cancer detection in prostate biopsies. Identification and characterization of additional methylation markers might further improve its high (80-90%) sensitivity. Studies have shown that methylation levels of GSTP1, APC, and RASSF1A are associated with advanced grade and stage. QMSP may augment the pathologic indicators currently used to predict tumor aggressiveness.
