Aims

We would like to used pooled reference samples to develop a sensitive magnetic bead-based assay to detect these glycoproteins with bisecting and core fucosylation changes. We have purified fusion-tagged glycoproteins with the specific glycan structures that we will spike into the control samples to determine the range and variation of detection prior to the phase II sample analysis. We will also measure the variance between the malignant ovarian cancer pooled samples and control benign pooled samples to determine if the assay can measure statistically significant differences.

Outcome

Three markers, periostin-LPHA, periostin-EPHA, and TSP1-AAL were evaluated in the BRSCW samples. Among the 94 controls we investigated if there were associations between markers and the following factors: age, race, menopausal status, parity, history of contraceptive use, smoking and hormone replacement therapy. There was only one association statistically significant between periostin-LPHA and HRT among post-menopausal women (p=0.04) and this may have occurred by chance given the number of associations examined. We compared the biomarkers measured in each case pool with the distribution of marker values in the corresponding control set. We found no suggestive results for periostin-LPHA or periostin-EPHA. For TPS1-AAL we found that the marker was elevated in the pooled sample from post-menopausal women with ovarian cancer and in the pooled sample from women with endometrial cancer. Again, given the number of associations examined these results may be due to random chance. Assay variability was high for all three biomarkers. Based on 16 replicates from a pooled control sample the co-efficient of variation was 110% for periostin-LPHA, 22% for periostin-EPHA, and 45% for TSP1-AAL.
